We see the [[SIGNATURE]]: Signature token in email notification creation of RCM module. Where do recruiter maintain signature so that it auto-populate in email notification when it triggers to candidate?

The [[SIGNATURE]] token pulls the signature (text only) configured in Admin Tools > E-Mail Notification Template Settings > Email Signature.

Enable this and update the content if needed.
